“利用反应扩散系统理论研究干细胞疗法治愈心力衰竭面临的瓶颈问题/ Theoretical study of the cause of induced arrhythmias after transplanting human pluripotent stem cell-derived cardiomyocytes for cardiac regeneration”
干细胞疗法被认为是治愈心肌梗死的可期新手段。最新研究却发现,尽管由多功能干细胞分化的心肌细胞(简称为PSC-CMs)能够修复诸如豚鼠等小型动物心肌受损部位,但推广到与人更相近的猕猴等大型动物时,反而会引起心律不齐等严重副作用。本项目旨在基于统计物理模型定量描述PSC-CMs的各项生理特征(如细胞膜电容量、电导系数、离子通道电流等)。接着利用基于反应扩散方程的大规模、高精度、并行化数值模拟,重点研究当注入大量生理特征不同于宿主原有心肌细胞的PSC-CMs后,大型动物的心肌及其受损部位会被引入何种程度的时空异构性,以及由此而来的大型动物更易诱发心律不齐的病因。进而探索防止或消除心律不齐的方法,为克服干细胞疗法在大型动物实验中产生的副作用提供理论指导,为随后的病患临床试验消除隐患。
Implanting human pluripotent stem cell-derived cardiomyocytes (hPSC-CMs) is intensively investigated as a promising approach in cardiac regeneration therapy. We are to study the realistic post-transplant cardiac tissue in which the differences of physiological properties between graft and host tissue are numerous and multiscale. The differences include but not are limited to conduction velocity, cardiomyocyte morphology, fiber anisotropy, ion channel current density, calcium handling and excitation-contraction coupling. These inhomogeneities existing in vivo may contribute to all fundamental arrhythmia mechanisms, namely automaticity, reentry, and triggered activity. To investigate mechanisms of inducing post-transplant arrhythmias under different conditions, we propose to establish a realistic cardiac model which is computationally efficient and sufficiently precise to theoretically study the arrhythmia initiation due to the transplantation through all physiologically relevant conditions of implanted inhomogeneities. Then, based on the understanding of mechanisms of arrhythmias, possible solutions in clinic would be proposed and tested in simulations to prevent this side effect in cardiac regeneration.
2. 学科教学(物理)/Physics Education
“基于学生课堂表情与增值性学习效果的关联性分析/correlation analysis of students’ facial emotion with value-added learning outcome”
高效课堂要求因材施教,即教师能够根据学生的反应(如表情等)实时地判断教学效果(即学生是否听懂)。这对于有效教学是十分重要的。如果缺乏这个互动过程,无论是课堂上缺乏经验的教师,还是现阶段慕课平台上的教学管理系统,就无法实时判断学生对当前知识点的学习认知情况,也就无法达到因材施教的教学效果。本研究旨在利用人工智能的人脸识别追踪与表情分析算法,实时定量测量课堂上每位学生的课堂表情值。同时,利用just-in-time教学法和Rasch模型构建适用于在线课堂和混合课堂的形成性评价体系,随堂评价每位学生的学习效果。最终利用大数据相关性分析理论定量研究什么样的学生表情对应于好的学习效果。基于研究结果我们将建立一整套利用学生表情实时反馈学习效果的智慧课堂系统,从而帮助课堂教师和慕课平台达到因材施教的高效课堂要求。
